Either way is fine since there is no tank hooked up, but we recommend having it turned off so that there is no chance of it unscrewing itself and falling off when you move around with it.
Sorry for the dumb question but when using the ASA Plug, would the Fill Adapter that the Plug is being used on be turned on or off? Amped Airsoft ASA Dual Tank Plug Adapter
